چکیده

1. The serum myeloma protein and urinary Bence-Jones protein of mice carrying the MPC-2 plasma-cell neoplasm have been purified by chromatography on ion-exchange diethylaminoethylcellulose and characterized. 2. The neoplastic plasma cells were found to form both proteins. Biosynthesis was studied by incubating sliced tumour tissue in vitro and following the incorporation of [14C]amino acids into the Bence-Jones protein and myeloma protein isolated by chromatography on diethylaminoethylcellulose and by precipitation with rabbit antisera. The time course of incorporation of radioactive amino acid into the two proteins is consistent with the view that Bence-Jones and myeloma proteins were formed independently by the neoplastic plasma cells and that neither protein was a precursor or breakdown product ofthe other. Approximately two molecules of Bence-Jones protein were formed for each molecule of myeloma protein. 3. The pool of extracellular plus intracellular protein associated with MPC-2 tumour tissue was determined by isotope-dilution techniques employing M3I-labelled Bence-Jones and 131I-labelled myeloma protein. About 3'6 mg. of myeloma protein and 0*4 mg. of Bence-Jones protein were found in 1 g. of tumour tissue (wet weight). This difference is probably caused by the rapid clearance of Bence-Jones protein from the plasma and extracellular fluid by excretion through the urinary tract.

Characterization of indole-positive Proteus mirabilis.

Thirteen indole-producing, swarming strains of Proteus were identified by additional biochemical testing as being Proteus mirabilis. These strains were characterized by 40 biochemical tests and by susceptibility testing to 11 antibiotics. All produced ornithine decarboxylase and were susceptible to members of the penicillin-cephalosporin groups of antibiotics. These indole-positive strains are ...

One-step biosynthesis of α-ketoisocaproate from l-leucine by an Escherichia coli whole-cell biocatalyst expressing an l-amino acid deaminase from Proteus vulgaris

This work aimed to develop a whole-cell biotransformation process for the production of α-ketoisocaproate from L-leucine. A recombinant Escherichia coli strain was constructed by expressing an L-amino acid deaminase from Proteus vulgaris. To enhance α-ketoisocaproate production, the reaction conditions were optimized as follows: whole-cell biocatalyst 0.8 g/L, leucine concentration 13.1 g/L, te...

The formation and development of large bodies in Proteus vulgaris OX-19. I. Bright phase contrast observations of living bacteria.

The appearance of diverse morphological forms in Proteus vulgaris OX-19 (Stempen and Hutchinson, 1951) stimulated interest in a cytological investigation of the organism to compare the chromatinic structures in the large bodies and other "atypical" forms with those in the "normal" rods. Cytological studies on large bodies of Proteus have been previously reported by KlienebergerNobel (1949), Tul...

Proteus vulgaris and Proteus mirabilis Decrease Candida albicans Biofilm Formation by Suppressing Morphological Transition to Its Hyphal Form

PURPOSE Candida albicans (C. albicans) and Proteus species are causative agents in a variety of opportunistic nosocomial infections, and their ability to form biofilms is known to be a virulence factor. In this study, the influence of co-cultivation with Proteus vulgaris (P. vulgaris) and Proteus mirabilis (P. mirabilis) on C. albicans biofilm formation and its underlying mechanisms were examin...

Methyl mercaptan and dimethyl disulfide production from methionine by Proteus species detected by head-space gas-liquid chromatography.

Head-space gas-liquid chromatography and mass spectrometry were used to detect and identify products formed by Proteus vulgaris, P. mirabilis, P. morganii, and P. rettgeri from a defined medium supplemented with either phenylalanine, methionine, valine, leucine, histidine, lysine, ornithine, threonine, asparagine, aspartic acid, or tryptophan. In a detailed study of the products formed by 68 st...
